BIO (6-bromoindirubin-3"-oxime) is a potent, reversible and ATP-competitive inhibitor for GSK-3α/β in the Wnt signaling pathway. BIO is the first pharmacological agent shown to maintain self-renewal in human and mouse embryonic stem cells¹. BIO-mediated activation of the Wnt signaling cascade maintained the undifferentiated state of both mouse and human embryonic stem cells under feeder free conditions possibly by Wnt-induced increase in the expression of Nodal^2-4.
